摘要
    耐高溫a一淀粉酶是目前最重要的工業(yè)酶制劑之一,約占酶制劑市場(chǎng)25%的比例。耐高溫a一淀粉酶能在較高溫度下能夠有效快速的分解淀粉,被廣泛應(yīng)用于紡織退漿、發(fā)酵、制糖等高溫條件下生產(chǎn)工業(yè)??莶菅勘U菌是國(guó)內(nèi)生產(chǎn)耐高溫a一淀粉酶的代表菌株之一。
    為了獲得較高的酶活和產(chǎn)量,在實(shí)驗(yàn)中,對(duì)耐高溫a一淀粉酶產(chǎn)生菌枯草芽孢桿菌的發(fā)酵培養(yǎng)條件進(jìn)行了優(yōu)化.綜合考慮了所有影響酶活的因素,并對(duì)培養(yǎng)條件進(jìn)行了統(tǒng)計(jì)分析,得到產(chǎn)耐高溫a一淀粉酶的最佳培養(yǎng)條件。結(jié)果表明:優(yōu)化培養(yǎng)基的組成(ZoomL)為玉米粉25,豆粉25,玉米漿10mL/L,CaCL2 4 Na2HPO4 3,檸檬酸鈉 3,PH6.8~7.0.該營(yíng)養(yǎng)條件下的酶活為2329u’mL一,,約是培養(yǎng)基優(yōu)化前酶活的12.6倍,酶活有了顯著的提高

ABSTRACT
Thermostable a-amysle, catalyzing hydrolysis of starch at high temperatures,is one of the most important .It is widely applied in desinzing,sugar and fermentation industry.bacillus subtilis is one of the industrial strains,which secretes thermostable a-amylase at high level
In the study ,the optimum medium for Bacillus subtilis .On the basis of single factor experiments,the prime factors affecting enzyme activity were selected.And then,the factors were optimized by response surface analysisThe result shows that the optimal medium composition was as follows:Corn flour25,Soybean meal25,Corn syrup 10mL/L, CaCL2 4, Na2HPO4 3,Sodium citrate3,PH6.8~7.0.Enzyme activity under the optimal culture conditions 2329 u/Ml.which is 12.6 fold of the orignai medium,this study has a significant advancement.The experimental data under various have validated the theoretical values
