摘 要 鱷嘴花是一種藥食兩用的植物,大量的文獻表明鱷嘴花具有抗癌,抗病毒,抗炎等多種藥理作用,但對其化學(xué)成分的研究尚未完整,為進一步了解鱷嘴花的活性物質(zhì),本課題組對鱷嘴花的化學(xué)成分及抗癌活性展開了研究。課題組在前期工作中,對鱷嘴花進行了系統(tǒng)分離,得到了乙酸乙酯、正丁醇兩個極性部位,并對乙酸乙酯部位經(jīng)粗分后得到了幾個流分。本人對這其中的兩個流分進行了進一步的分離純化,制備單體,并對單體進行結(jié)構(gòu)鑒定,確定所得到化合物的結(jié)構(gòu)。主要研究工作及結(jié)果如下:
(1) 通過色譜分離技術(shù)如常規(guī)的薄層層析摸索分離條件,利用正相硅膠柱,ODS柱等柱層析對對乙酸乙酯部位提取物進行分離提純,再進一步用重結(jié)晶等方法進行純化,制備單體化合物,分離純化出化合物1及2。
(2) 利用HPLC為檢測手段測定化合物1及2純度,它們的HPLC結(jié)果顯示純度均達到99%以上,由此確定化合物1和2已經(jīng)達到進行波譜分析的要求。
(3) 將純度達標(biāo)的單體化合物1進行波譜測定,質(zhì)譜顯示化合物1的分子量為427.40,結(jié)合1H-NMR、13C-NMR、DEPT、HSQC等波譜技術(shù),通過查閱和對比文獻確定化合物1為羽扇豆醇。
(4) 將純度達標(biāo)的單體化合物2進行波譜測定,通過MS確定分子為388.60,但其具體的結(jié)構(gòu)仍在分析當(dāng)中。

Separation, purification and identification of Polyterpenoids of
Clinacanthus nutans(burm.F)Lindauthe
Abstract Clinacanthus nutans(burm.F)Lindau is a kind of medicinal and edible plant, a lot of literatures show that Clinacanthus nutans(burm.F)Lindau with anti-cancer, anti-viral, anti-inflammatory and other pharmacological effects, but the study of its chemical composition is not yet complete, in order to further understand the active substance of Clinacanthus nutans(burm.F)Lindau , The research group carried out studies on the chemical constituents and antitumor activity by Clinacanthus nutans(burm.F)Lindau. The Group in the previous work, the Clinacanthus nutans(burm.F)Lindau were isolated, the ethyl acetate, n-butanol two polar fractions, and the ethyl acetate fraction rough points obtained several stream. I did for the two fraction was purified further separation, preparation of monomer, and the structures were identified on the monomer, determine the structures of the compounds. The main research work and results are as follows:
(1) By chromatographic separation techniques such as TLC conventional fumble separation conditions, using normal phase silica gel column, ODS column chromatography of the extract of ethyl acetate fraction was isolated and purified by recrystallization, and then further purified preparation methods, compounds, separation of pure compounds 1 and 2 respectively.
(2) The compound 1 and 2 detection means purity using HPLC for the determination, The HPLC results showed that the purity reaches more than 99%, the compounds 1 and 2 have reached the requirements for spectrum analysis.
(3) The compounds 1 in the purity of standard wave spectroscopy, mass spectrometry showed that molecular was 427.40, according to 1H-NMR, 13C-NMR, DEPT, HSQC spectroscopy, by consulting literature and comparison to determine the compound 1 as lupeol.
(4) The compounds 2 in the purity of standard spectroscopic measurement, determined by MS molecule is 388.60, but the structure is still in analysis.
