摘要:本文以韭蘭為材料,對植物染色體常規(guī)壓片,去壁低滲法制片技術(shù)、染色效果進(jìn)行了比較。實(shí)驗(yàn)表明:去壁低滲法獲得細(xì)胞和染色體都較分散,Giemsa染色使得背景與材料的對比度較大,便與觀察。不足之處在于去壁低滲過程中,時(shí)間特別不易掌握,耗材多,且成功率較低。染色體容易丟失、形態(tài)也不太理想。常規(guī)壓片耗材少,成功率較高,但不易做分帶制片。
